Step 1: Creating Channels
- To perform the experiment, you need the Physiology (Dynamics) module.
- You have a Sutter DG4/5 with appropriate excitation filters for Fura-2 and a Fura-2 filter set in the microscope's reflector wheel.
- You are on the Acquisition tab.
- Create a new experiment in the Experiment Manager, e.g. "Physiology Fura-2".
- Add the channel Fura-2 using Smart Setup.
- Activate the Time Series checkbox in the acquisition dimensions.
- Open the Channels tool.
- Select the Fura-2 channel from the list.
- Click on Options
and select Duplicate.
- Select the first Fura-2 channel from the list.
- Click on the Options
and select Rename.
- You can now rename the channel, e.g. Fura-2 340 nm.
- Repeat steps 7 and 8 to rename the second channel, e.g. Fura-2 380 nm.
- Select the Fura-2 380 nm channel.
- Select another LUT from the dropdown list, e.g. red.
- Select the entry 21 HE Ex. FURA 380 from the Excitation dropdown list.
- The excitation filter is used for this channel.
- Adjust the exposure time and focus for both channels.
- You have created the channels for your experiment.
