Customizing a Reference Scan Profile
- You have licensed the Spatial Biology toolkit and activated it in Tools > Toolkit Manager.
- You have a reference scan profile that was set up by an expert user.
- In the Magazine view, click Customize Reference Scan Profile.
- The first step of the Customize Reference Scan Profile wizard opens.
- For Reference Scan Profile, select the scan profile you want to adapt, and define a Batch Name.
- The scan profile is selected.
- Click Next.
- The next step opens.
- Check all the listed channels from the original scan profile and activate the checkbox for all channels you want to use. If necessary, adapt the focus strategy and marker.
- You have reviewed and selected channels for the adapted profile.
- Click Next.
- The next step opens.
- Select the reference slide with the two drop-downs and click Onboard Slide. Make sure you select a slide that is typical for the current batch of slides you want to scan.
- The reference slide is loaded and an overview experiment is executed, including the tissue detection.
- The acquired image is displayed in the Center Screen Area.
- Use the controls to define the exposure times for the individual channels. Click Auto Exposure to start an exposure time measurement for all channels, or click Auto for a single table entry to start a measurement for that respective channel. Alternatively, set the exposure time manually with the input field in the table.
- The exposure times are set.
- Click Snap to acquire a snap, or click Live to start a live acquisition.
- An image is displayed in the Center Screen Area.
- Check the image to see if the exposure times are set correctly. If necessary, repeat the previous steps to further adapt the exposure times.
- You have checked the updated exposure times.
- To save the values when executing multiple exposure time measurements, click Record Exposure Times before a new measurement or exposure time change.
- The exposure times for each marker are saved and displayed in the table of the Recorded Exposure Times section.
- When you have set suitable exposure times for the new batch of slides, click Finish.
- All changes are saved and the wizard closes.
- The reference scan profile is adapted and a copy of the original scan profile is created.
