Step 1: Selection of Autofluorescence Spectra
With this step you select autofluorescence spectra. You have the following possibilities:
- Load an xml file which defines the autofluorescence spectrum, for example a file from a previous run of this wizard.
- Load an image and draw graphical elements into the image which indicate autofluorescence spectra and background.
If you load an image, it is displayed in the Center Screen Area and you have the 2D and Split view available. If you load an xml file, the chart of the spectrum is displayed in the Center Screen Area.

Show Spectrum |
Only visible if you have selected autofluorescence and background with the drop-down below. Activated: Displays a chart with the currently selected autofluorescence spectrum in the Center Screen Area. |

Select Autofluorescence |
Only visible if you have loaded a czi file and you have drawn a graphic into the image. Selects the graphical element that marks the autofluorescence with the drop-down list. |

Select Background |
Only visible if you have loaded a czi file and you have drawn a graphic into the image. Selects the graphical element that marks the background with the drop-down list. If there is no background in the image, select None. |

Fixed Background |
Only visible if Select Background is set to None and if the necessary information are not present in the file metadata. Sets a fixed value for the background. In case of HDR scans the value should be set to 255, in other cases to 64. |
