Examples for Custom Features

The following examples illustrate the functionality of creating custom features in the image analysis wizard, see also Creating Custom Features.

Example 1: Number of Spots per Cell

This custom feature calculates the number of spots in each cell and is based on an analysis setup for an image with cells (DAPI) and spots (EGFP). The class tree could look like this:

To create this custom feature you have to define the following operand and expression for the class Cell:

  • Operand a:
    • Class: All Spots per Cell
    • Feature: Count
  • Define Custom Expression: a

The subsequent run of the analysis setting will add your custom feature as another column to the analysis table for the class Cell.

Example 2: Average Area per Spot

This custom feature calculates the average area per spot in a cell by dividing the area of the cell through the number of spots in that cell. It is based on an analysis setup for an image with cells and spots. The class tree could look like this:

To create this custom feature you have to define the following operands and expression for the class Cell:

  • Operand a:
    • Class: Cell
    • Feature: Area
  • Operand b:
    • Class: All Spots per Cell
    • Feature: Count
  • Define Custom Expression: a/b
